Lena Endre planerade först att bli marinbiolog men hoppade av studierna. 1979 började hon spela amatörteater i den fria gruppen Teater Sputnik och vid Inge Waerns teaterstudio. 1983 kom hon in på Scenskolan i Stockholm där hon bland annat var klasskamrat med Gunilla Röör.
