His father was a pharmacist and his mother a kindergarten teacher. She also played piano and sang, and introduced Tommy to opera and big band music on the radio.

Henderson served as the original bandleader for The Tonight Show with founding host Steve Allen (as well as for Allen's Sunday-night variety show), then came back to Tonight after the departure of host Jack Paar and his orchestra director José Melis.
He was best known for his longtime association with The Tonight Show Band, led by Doc Severinsen, on the television program The Tonight Show Starring Johnny Carson. He was a member of the band from 1971 until Carson's retirement in 1992.
