Aug 2, 2018 · Morphology: G. lamblia exists in two morphological form- trophozoite and cyst. Trophozoite: It is the active feeding stage of parasite which is responsible for colonization in intestine. The shape of trophozoite is pear shape or tennis racket shape with broad round anterior end and a tapering posterior end.
Giardia duodenalis cysts are oval to ellipsoid and measure 8-19 µm (average 10-14 µm). Mature cysts have 4 nuclei, while immature cysts have two. Nuclei and fibrils are visible in both iodine-stained wet mounts and trichrome-stained smears.
Sequence analysis revealed a cysteine-rich (12%) protein with frequent CXXC motifs ( 6 ). Subsequent sequences of a number of vsp genes have shown a number of characteristics common to all vsp genes. They are all cysteine rich (12%), and most cysteines are present in CXXC motifs.

Sometimes the correct identification of this morphological character may need to the examination of the permanent stained smear for revealing some details of the organism that cannot be seen in unstained or temporary stained smears.
