He was Professor of Comparative Zoology at the University of Catania from 1883, and Professor of Comparative Anatomy at Sapienza University of Rome from 1895 until his death.

The son of Luigi Grassi, a municipal official, and of Costanza Mazzuchelli, a peasant of unusual intelligence, Grassi was educated at Saronno. From 1872 he studied medicine at Pavia, graduating in 1878.
