Entamoeba cells are small, with a single nucleus and typically a single lobose pseudopod taking the form of a clear anterior bulge. They have a simple life cycle. The trophozoite (feeding-dividing form) is approximately 10-20 μm in diameter and feeds primarily on bacteria.
Its body is covered by plasma lemma and cytoplasm is differentiated into ectoplasm and endoplasm. There is a single large, broad and blunt pseudopodium formed of ectoplasm. Endoplasm contains single spherical nucleus and food vacuoles. Nucleus has peripheral crown of chromatin blocks and a centrally located nucleolus.

Mature Entamoeba histolytica/Entamoeba dispar cysts have 4 nuclei that characteristically have centrally located karyosomes and fine, uniformly distributed peripheral chromatin. Cysts usually measure 12 to 15 µm.
Entamoeba hartmanni, also in the quadrinucleate cyst clade, is much smaller than E. histolytica with cysts reaching only 10 μm in diameter and trophozoites of 3–12 μm in diameter.
