The 'Pampanito', the sub used in the movie, is a museum at pier 45 in San Francisco. Commander Dodge makes a snarky comment before meeting his crew, if they're leftovers from World War II. His Chief Engineer, Howard, is played by Harry Dean Stanton. Stanton served as a cook on board the USS LST-970 during the Okinawa Campaign in World War II.
Much of Up Periscope is filmed on a real submarine at sea, indicating full Navy cooperation. This may explain why the script is lacking in dramatic tension -- no matter what happens, Navy policy or prestige is never in question.

Up Periscope was released in '59 (1959). Many years later a parody of the submarine movie genre would be made both in a year with a reverse of this film's year of release (i.e. in '95 (1995)) and with also a title that reverses this movie's title (i.e. Down Periscope ).
Lieutenant Braden discovers that Sally, the woman he's been falling in love with, has actually been checking out his qualifications to be a U.S. Navy frogman.
