Nov 15, 2022 · Abstract. Selenocysteine (Sec), the 21st genetically encoded amino acid, is structurally similar to cysteine (Cys) but with a sulfur to selenium replacement. This small change confers Sec with related chemical properties to Cys but often with enhanced reactivity. In organisms, Sec is present in selenoproteins taking on various roles such as ...

Oct 9, 2012 · The mechanism of selenocysteine synthesis. Selenocysteine, also known as the 21st amino acid, is unique among the proteinogenic amino acids. It is the only amino acid containing an essential dietary micronutrient (selenium) as a constitutive component, the only amino acid encoded by a UGA codon and the only one synthesized on its tRNA in all domains of life.

Selenocysteine (symbol Sec or U, [4] in older publications also as Se-Cys) [5] is the 21st proteinogenic amino acid. Selenoproteins contain selenocysteine residues. Selenocysteine is an analogue of the more common cysteine with selenium in place of the sulfur . Selenocysteine is present in several enzymes (for example glutathione peroxidases ...

Aug 1, 2013 · Abstract. Selenium is an essential trace element that is incorporated into 25 human proteins as the amino acid selenocysteine (Sec). The incorporation of this amino acid turns out to be a fascinating problem in molecular biology because Sec is encoded by a stop codon, UGA. Layered on top of the canonical translation elongation machinery is a ...

thesis . A selenocysteine residue is encoded by the UGA codon in selenoprotein mRNA , which is decoded by selenocysteyl-tRNA [Ser]Sec [ 3 , 4 ]. The selenocysteine moiety on selenocysteyl-tRNA [Ser]Sec is biosynthesized using selenophosphate , which is produced from selenide and ATP by selenophosphate synthetase [ 5 , 6 ].
Over the years, the molecular mechanism that allows the UGA-misinterpretation for Sec incorporation was identified as being dependent on a specific mRNA element namely SECIS (SElenoCysteine Incorporation Sequence). This unique sequence in the mRNA folds in a hairpin conformation downstream, in Bacteria, or at the 3′ untranslated region–3 ...
Given the rapid progress in the field of selenocysteine biosynthesis, we should not have to wait long for answers to the new questions. References Palioura, S. et al. Science 325 , 321–325 (2009).
