  1. Oct 9, 2012 · The mechanism of selenocysteine synthesis. Selenocysteine, also known as the 21st amino acid, is unique among the proteinogenic amino acids. It is the only amino acid containing an essential dietary micronutrient (selenium) as a constitutive component, the only amino acid encoded by a UGA codon and the only one synthesized on its tRNA in all domains of life.

  2. Nov 15, 2022 · In bacteria, Ser-tRNA Sec is converted to Sec-tRNA Sec in a single step by selenocysteine synthase (SelA). In eukaryotes this process occurs in two separate steps: i) phosphorylation of Ser by O -phosphoseryl-tRNA kinase (PSTK) then ii) conversion of phosphoserine (Sep) to Sec by Sep-tRNA:Sec-tRNA synthase (SepSecS).

  3. Mar 10, 2011 · The complete Sec biosynthetic pathway. The complete biosynthesis of Sec on its tRNA is shown in Figure 1. Initially, tRNA [Ser]Sec is aminoacylated with serine by SerS ( 17) and the seryl moiety is then converted to O -phosphoseryl-tRNA [Ser]Sec by PSTK ( 15 ).

  5. Aug 1, 2013 · Abstract. Selenium is an essential trace element that is incorporated into 25 human proteins as the amino acid selenocysteine (Sec). The incorporation of this amino acid turns out to be a fascinating problem in molecular biology because Sec is encoded by a stop codon, UGA. Layered on top of the canonical translation elongation machinery is a ...

  6. thesis . A selenocysteine residue is encoded by the UGA codon in selenoprotein mRNA , which is decoded by selenocysteyl-tRNA [Ser]Sec [ 3 , 4 ]. The selenocysteine moiety on selenocysteyl-tRNA [Ser]Sec is biosynthesized using selenophosphate , which is produced from selenide and ATP by selenophosphate synthetase [ 5 , 6 ].

  7. Jun 12, 2018 · Selenium (Se) is an essential trace element for several organisms and is mostly present in proteins as l-selenocysteine (Sec or U). Sec is synthesized on its l-seryl–tRNASec to produce Sec–tRNASec molecules by a dedicated selenocysteine synthesis machinery and incorporated into selenoproteins at specified in-frame UGA codons. UGA–Sec insertion is signaled by an mRNA stem-loop structure ...

  8. Jan 22, 2020 · The initial Ser residue present in the Ser-tRNA [Ser]Sec is phosphorylated by PSTK, resulting in an intermediate o-phosposeryl- (Sep)-tRNA [Sep]Sec. Following the conversion, the o-phosphoseryl-tRNA Sec selenium transferase (SepSecS–a PLP-dependent enzyme) performs the Sep/Sec conversion, resulting in the mature Sec-tRNA Sec ( Liu et al., 2014 ).
