E. coli has cysts in size up to 10 to 35 micrometers, the shape is irregular, oval with a shell-like appearance that is more uniformed compared to E. histolytica, and has up to eight nuclei in the cyst compared to the four nuclei of E. histolytica.

Entamoeba histolytica and Entamoeba dispar are morphologically identical species. In bright-field microscopy, E. histolytica/E. dispar cysts are spherical and usually measure 12 to 15 μm (range may be 10 to 20 μm). A mature cyst has 4 nuclei while an immature cyst may contain only 1 to 3 nuclei.

The cysts of Entamoeba histolytica, shown below, measure between 10 and 20 µm in diameter. The key identification feature is the number of nuclei; cyst will have 1 to 4 nuclei. In addition, the peripheral chromatin is generally evenly distributed. In the images below, two nuclei are clearly visible.
