Synthesis of PCP from PCA via 1-phenylcyclohexanol (PCOH) The PCOH starting material can be purchased or prepared by reaction of phenyllithium or phenyl grignard with cyclohexanone. This alcohol is then be transformed into the amine by reaction with sodium azide (~90% yield) followed by reduction.
Example: Synthesis of PCE (N-ethyl-1-phenylcyclohexylamine): A mixture of 100 g of anhydrous ethylamine and 220 g of cyclohexanone was kept 16 hours, then shaken with solid KOH. The lower oil layer, which comprises the imine ( N-cyclohexylidenethylamine), was then removed by decantation.
A mixt. of 190 g. p-MeC6H4SO3H.H2O in 250 ml. PhMe is heated under a H2O trap until anhyd. A soln. of 165 g. I in 500 ml. Et2O is added to this anhyd. soln. at 0-5 Deg, followed by a soln. of 1 mole PhMgBr in 750 ml. Et2O with stirring 30 min. at 5 Deg, the mixt. decompd. by the addn. of excess satd. NH4Cl and concd.
